PROCESS OVERVIEW

Througout the entire production process, product in tanks is easily tested for the presence of spoilage bacteria and wild yeast using FastOrange bouillon broth products.

Combine finished product in a 1:1 ratio with FastOrange B (2036-1) or FastOrange Wild Yeast  (2039-1and incubate.

Contaminated product will change the media color from violet to yellow and may form sediment or haze.

ESSENTIAL

Follow ASEPTIC SAMPLING TECHNIQUE  when collecting samples and create a STERILE FIELD when performing the testing steps below.

EQUIPMENT*

Needed to Collect Samples

  1. Spray or wash bottle of 70% isopropyl alcohol
  2. Sterile swabs or nylon brush
  3. Butane torch
  4. Sterile flask or Whirl-Pak with volume of > 200 ml
 

STEP 1

Collect Samples

-Follow the SOP for collecting aseptic samples using a sample valve.

STEP 2

Prepare Work Area

-Follow the SOP for preparing the work area for sample processing.

EQUIPMENT*

Needed to Process Samples

  1. Spray or wash bottle of 70% isopropyl alcohol 
  2. Clean paper towels
  3. Bunsen burner or lamp
  4. Sample flask or Whirl-Paks
  5. Appropriate FastOrange media

STEP 3

Prepare the Sterile Area

-Follow the SOP for preparing a sterile area.

STEP 4

Prepare the Medium

-Working inside the sterile area…

-Pipette or pour 50 ml of 2036 or 2039 media into the number of sterile flasks or whirl-paks that equal the number of tests being performed

-Close or cover media containers

-Re-seal and set FastOrange media bottles aside.

STEP 4

Combine Product and Medium

-Working inside the sterile area

-Gently swirl the first sample container to re-suspend any sediment that may have precipitated out during storage

-Pour or pipette 50 ml of re-suspended product into the first FastOrange flask, bottle, or whirl-pak

-Close or seal containers with sample + medium mixture

NOTE: If using Whirl-Paks, push as much air as possible out of the bag before sealing the top.

-Repeat if testing the same sample across multiple FastOrange media, if running duplicates of the same medium, or if processing samples from multiple tanks

-Extinguish the Bunsen burner and clean the sample processing area.

STEP 5

Incubate

-Confirm that all flasks and/or whirl-paks are well-sealed.

-Place upright in a warm area or incubator

-To prevent spills or leaks, place flasks or paks inside a shallow pan or bucket before heating

-Incubate at 25 ± 2 °C (77 ± 4 °F) for 3-7 days
